研究方向:DNA是主要的遗传物质。在真核细胞中,在维持转录和复制等重要生物学功能的同时,线性长度可达数米的基因组DNA能够被高度折叠包裹在直径只有几微米的细胞核中。对细胞核内基因组的研究是理解遗传信息的表达和传递的关键。本课题组致力于发展在单细胞中探索基因组结构和功能的测序技术,主要包括全基因的精确测量,染色体三维空间结构的重构,染色质开放区域的观测,核小体的定位以及蛋白质与DNA的相互作用的测定等方面。并以此为核心对基因组稳定性、基因表达调控和基因组的三维结构与功能等领域的基本生物学问题展开深入研究。同时在此基础上深入探索与基因组异常相关的重大疾病的分子机理和潜在的治疗策略。
